If I connect my iPad to my iPhone hotspot, will I be able to change the home location on the fly?

You will not be able to change the home point to your location. You could change it to the Phantom's current location though (even without the hotspot).

When connected to a hotspot, only the Internet connection is shared. It's not possible to share GPS data.
